Eikon Therapeutics Eyes 2027 Melanoma Trial Update, With Key Cancer Data Expected in 2029

Key Points

  • Eikon expects an interim analysis of its lead melanoma trial in the first half of 2027, with definitive results from the melanoma and lung cancer studies potentially arriving around mid-2029. Timing depends on trial events, and the company remains blinded to treatment-group results.
  • Early lung cancer data for EIK1001 showed encouraging response rates and duration, but came from a single-arm study and do not establish that the drug improves outcomes. The FDA said similar results in a randomized trial could prompt an accelerated-approval evaluation.
  • Eikon is advancing selective PARP1 inhibitors designed to limit blood-cell toxicity, with studies exploring combinations with chemotherapy and other cancer treatments. It also reported preliminary findings from an experimental Werner helicase inhibitor, which the company cautioned do not establish efficacy.

Eikon Therapeutics NASDAQ: EIKN expects an interim analysis of its lead melanoma study in the first half of 2027 and estimates that definitive data from that trial and a lung cancer study could be available around the middle of 2029, CEO Roger Perlmutter said during a JPMorgan biotech call.

Both studies are testing EIK1001, an experimental immune-stimulating drug, alongside standard cancer treatments. Perlmutter cautioned that the timing of the analyses depends on events such as disease progression, and that the company remains blinded to results by treatment group.

Eikon has roughly 1,000 patients enrolled across oncology trials, Perlmutter said. He expects additional data over the next six to 12 months from programs spanning melanoma, lung, breast, ovarian and prostate cancers, as well as tumors with defects in DNA repair.

Lead Cancer Trials Advance

EIK1001 activates the toll-like receptors TLR7 and TLR8, an approach intended to stimulate an immune response against tumors. The registration-enabling melanoma and non-small cell lung cancer studies compare the drug plus standard care with placebo plus standard care.

Perlmutter said the melanoma study has advanced further. An independent monitoring committee recommended a dose of 0.6 milligrams per square meter after comparing it with a higher dose. The next interim analysis will assess whether the study should proceed into its larger phase 3 portion; it is not powered to establish a treatment effect, he said.

The lung cancer trial will undergo a dose-selection step followed by an event-driven analysis. Perlmutter said he expects the two programs to reach definitive results at roughly the same time, although the 2029 estimate is uncertain.

Eikon presented mature phase 2 data at the ESMO cancer meeting for patients with squamous non-small cell lung cancer who received EIK1001 with Merck’s Keytruda and chemotherapy. Perlmutter said the results showed a higher-than-expected response rate and lengthy responses, but emphasized that the single-arm study cannot be directly compared with other trials.

He said the Food and Drug Administration agreed that the data supported a registration-enabling trial and told Eikon to request an accelerated approval evaluation if similar response and duration results appear in the randomized study. That possibility depends on future data; Perlmutter said randomized results are needed to establish whether EIK1001 improves outcomes.

Systemic activation of TLR7 and TLR8 can cause cytokine release syndrome, a potentially serious inflammatory reaction. Perlmutter said Eikon has treated more than 600 patients with EIK1001 and uses body-surface-area dosing, among other measures, to manage the risk. He described the observed rate of grade 1 or 2 cytokine release syndrome as being in the single digits.

PARP Program Targets Combination Use

Eikon is also developing two selective PARP1 inhibitors, EIK1003 and EIK1004. Perlmutter said the company designed the program around preclinical evidence that inhibiting the related PARP2 enzyme contributes to blood-cell toxicity seen with existing PARP inhibitors, though he cautioned that the mechanism has not been established in humans.

Among the first 68 patients whose EIK1004 data were presented at ESMO, one had grade 3 anemia, he said. He reported a single-digit rate for EIK1003 and said the drugs’ tolerability could allow them to be combined with chemotherapy earlier in treatment.

Eikon presented EIK1003 data in combination with paclitaxel in previously treated patients. Perlmutter said the majority had either tumor responses or stable disease. He also cited a roughly 15% transfusion rate in a study combining EIK1003 with abiraterone in heavily pretreated prostate cancer patients.

The company has begun studying EIK1003 with platinum chemotherapy and a taxane, a combination that could support first-line use. Perlmutter said Eikon is considering several registration opportunities and has not made a final decision, although a maintenance-therapy study is unlikely to be its first priority because it could take longer to produce results. EIK1004, which is designed to penetrate the central nervous system, may be of particular interest in patients prone to brain metastases, he said.

Earlier-Stage Pipeline

Perlmutter also discussed an experimental Werner helicase inhibitor developed using Eikon’s microscopy platform. In initial data from 10 cancer patients, all six with microsatellite-unstable tumors had stable disease on the drug, he said. A separate patient receiving the drug with Keytruda had a partial response. He stressed that the findings were early and did not establish efficacy.

Eikon has also filed an investigational new drug application for EIK1006, an experimental androgen receptor antagonist, and expects to begin enrolling patients soon, Perlmutter said.
